Yes, you have to open the reserve (lower the lever) or the tank will not fill.
The mechanism in most reserves is like a one way valve with a spring pushing a plunger that restricts the flow of air when the pressure is low. Lowering the lever retracts the one way valve plunger.
When you go to fill the tank if the one way valve plunger is not retracted it will not allow air into the tank.

Since the reserve mechanism operates by rotating 90°, some divers used to rig them so pushing up opened the reserve. The thought was that this made it less likely to pull the rod while swimming through kelp. I think it just confused the hell out of people.
Some of my customers have requested that I remove the reserve mechanism from their valves. Now they can fill with the lever up or down.
Listen for air entering cylinder, or if filling off a small compressor, check cylinder pressure after filling gauge indicates proper pressure. If no air is heard going in or cylinder isn't filled, flip the reserve to other position.
In 99.9% of cases what luis said will be the case, so always start in the down position.
